Q: Does this use an app ?

No u don’t need a app. They have 10 year battery in them

Q: Would high humidity set these off? Want for a fish tank room

It needs water to make the connection and set the alarm off. I have mine under the sink units where the water would goto if there was a leak. If in the high humidity there was condensation on the devices then I think they would alarm. It’s that much condensing humidity you kind of have a leak and would need to control the humidity. It will not work if completely submerged, however, it should float if the water gets to that level. There are other products on the market which will connect to you phone & alert you directly if away at any time. I have these installed as well, for precaution. Trust this helps

Q: Does this product work with reverse osmosis (ro) water, which has a significantly lower level of conductivity than standard tap water?

As it works on resistance I doubt it will work with RO , however the RO will pick up impurities as it runs along a surface so depends on your use case .
