Q: I bought a pair of these, and used them twice, when once of the speakers now fails to power up. Is there a replaceable fuse?

I believe so! Unplug the power cord and lift the flap, the fuse should be in there.

Q: Can these speakers be primary for Dj and karaoke

Yes you can use the speakers for DJ and especially karaoke there is more then one microphone plugin outlets

Q: would you need another powered speaker to connect to the 1/4" line out to act as a monitor? or can you just use a non powered/passive speaker?

I do believe that you can use a passive speaker with the 1/4 inch but I've never done it.
