Yes one size. Shouldn't be a problem as it basically just wraps around the bar so doesn't really matter what size your hands are. Hope this helps.

Have any women used these and do they actually protect against calluses?

They stretch so would be suitable as unisex and the definitely protect the hand from calluses. They also help your grip, meaning you can lift a bit more before you hands exhaust.

Are these real leather?

Its only the palm and Im not 100% sure if its leather or synthetic but I liked mine enough to reorder. Once they have softened up they are so much better than gloves, rarely recommend anything but not just for the price but for the durability you cant lose with them.
