What is the construction? Is it Aluminum or painted steel?

It is anodized aluminum in black. The base is a heavy cast aluminum. The biggest problem is that their fastening system stinks. But it's an easy fix, using lock washers and flat washers. I also changed the Allen screws with torx head. If you are using old type monitors that are heavy. You'll need to remove the plastic washers they use on the monitor brackets and use metal with lock washers. Making sure you tighten them as tight as possible. Otherwise they will keep falling. The unit itself is excellent, and made of good material. They just cheaped out on the fasteners. Like. Stated it's a great cart to use if you need mobility and don't have a lot of room. To fix the fastener problem cost me about $10.00 and an hour then remove and reinstall everything. But since you are now aware of this and if you do buy it. Just get the fasteners beforehand. Are the printer and tower holders interchangeable?

Yes you can move the aroun to a position that works best for you. But I'd you are asking can you put your tower on the printer, platform not really. The tower platform is able to clamp the tower down so it doesn't fall. But as far as configuring them, so they work for your application yes they are

Are the shelving positions flexible? Can all shelves be mounted on one side?

they can all be installed on the front , but on side all but monitor/keyboard shelves because they have larger stabilizing brackets
