We too often have six-players. Can 1 core set + 2 hero character packs work for six players or do I need 2 core?

You need two base sets or you can play the 6 player team play variant included in the base rules. With all 5 character packs you could play 5 players with the base set or add a 6th boss deck. Honestly though, most people would agree the game plays best at 2 and every player beyond that is less recommended.

No, the core deck will support 2-4 Players.

No. Those are separate cards available on the White Wizard website. There's five packs each with a different class.
